    Posterior Reversible Encephalopathy Syndrome

    Background: The presentation of posterior reversible encephalopathy syndrome (PRES) features neuropsychiatric symptoms in the context of predominantly white matter cerebral edema in the setting of a diverse variety of underlying clinical entities. Objective: To illustrate the presentation and diagnostic strategy for this under-recognized condition. Method: We present two cases of PRES and review the available literature. Results: PRES may be due to a number of underlying conditions, but typically presents with symptoms consistent with delirium. Conclusions: Psychiatrist practicing in the general hospital should be aware of the presentation and appropriate work-up of PRES to forestall serious potential sequelae

    Fandom and activism: Experimenting with memetic communication appeals about human rights issues during the 2022 winter Olympic games

    In the sports context, activism is a popular focus of inquiry. In fact, scholars have posited that sports present an ideal platform for human rights and political activism and protest (Agyemang, Singer, & DeLorme, 2010; Coombs & Cassilo, 2017; Kaufman & Wolff, 2010). This study aims to analyze the persuasiveness of memetic communication in the global sports context by investigating the influence of exposure to social advocacy memes on issue importance and general attitudes toward global human rights. This experiment relied on MTurk participants in an online experiment using Qualtrics software hosted by the Communication Research Lab at a Midwest University. We measure the persuasive influence of a variety of persuasive strategies (e.g., assertive, comparative, calls to action, and appeals to self-efficacy) as well as explicit affiliations within a real-time global sports context (e.g., 2022 Winter Olympics). These findings add to existing scholarship by focusing on post-exposure behavioral intentions (e.g., diffusion intentions, advocacy intentions, consumption intentions, and information-seeking intentions) likely to spur activism and advocacy. These findings support the position that memetic communications help to serve as a motivational catalyst for information-seeking behaviors about global human rights issues in real-time global sports contexts.     Analyzing the Prophylactic and Therapeutic Role of Inoculation to Facilitate Resistance to Conspiracy Theory Beliefs

    Conspiracy theories pose a variety of social and psychological consequences for individuals and society, and research suggests that around half of the U.S. population believes at least one. A two-phase inoculation experiment was conducted. Inoculated participants reported more negative general attitudes toward conspiracy theories and lower Phase II generic conspiracist beliefs, which are both indicators of harm-reduction and the beneficial healing impacts of therapeutic inoculation. The addition of therapeutic inoculation as a harm-reduction or healing technique in practitioners’ stakeholder response toolkit is a valuable contribution to both theory and practice

    Human Sulfatase 2 inhibits in vivo tumor growth of MDA-MB-231 human breast cancer xenografts

    BACKGROUND: Extracellular human sulfatases modulate growth factor signaling by alteration of the heparin/heparan sulfate proteoglycan (HSPG) 6-O-sulfation state. HSPGs bind to numerous growth factor ligands including fibroblast growth factors (FGF), epidermal growth factors (EGF), and vascular endothelial growth factors (VEGF), and are critically important in the context of cancer cell growth, invasion, and metastasis. We hypothesized that sulfatase activity in the tumor microenvironment would regulate tumor growth in vivo. METHODS: We established a model of stable expression of sulfatases in the human breast cancer cell line MDA-MB-231 and purified recombinant human Sulfatase 2 (rhSulf2) for exogenous administration. In vitro studies were performed to measure effects on breast cancer cell invasion and proliferation, and groups were statistically compared using Student's t-test. The effects of hSulf2 on tumor progression were tested using in vivo xenografts with two methods. First, MDA-MB-231 cells stably expressing hSulf1, hSulf2, or both hSulf1/hSulf2 were grown as xenografts and the resulting tumor growth and vascularization was compared to controls. Secondly, wild type MDA-MB-231 xenografts were treated by short-term intratumoral injection with rhSulf2 or vehicle during tumor growth. Ultrasound analysis was also used to complement caliper measurement to monitor tumor growth. In vivo studies were statistically analyzed using Student's t test. RESULTS: In vitro, stable expression of hSulf2 or administration of rhSulf2 in breast cancer cells decreased cell proliferation and invasion, corresponding to an inhibition of ERK activation. Stable expression of the sulfatases in xenografts significantly suppressed tumor growth, with complete regression of tumors expressing both hSulf1 and hSulf2 and significantly smaller tumor volumes in groups expressing hSulf1 or hSulf2 compared to control xenografts. Despite significant suppression of tumor volume, sulfatases did not affect vascular density within the tumors. By contrast, transient exogenous treatment of MDA-MB-231 xenografts with rhSulf2 was not sufficient to inhibit or reverse tumor growth. CONCLUSION: These data indicate that in vivo progression of human breast cancer xenografts can be inhibited with sulfatase expression, and therapeutic effect requires constant delivery at the tumor site. Our results support a direct effect of sulfatases on tumor growth or invasion, rather than an effect in the stromal compartment

    A comprehensive study of GRB 070125, a most energetic gamma ray burst

    We present a comprehensive multiwavelength analysis of the bright, long duration gamma-ray burst GRB 070125, comprised of observations in γ\gamma-ray, X-ray, optical, millimeter and centimeter wavebands. Simultaneous fits to the optical and X-ray light curves favor a break on day 3.78, which we interpret as the jet break from a collimated outflow. Independent fits to optical and X-ray bands give similar results in the optical bands but shift the jet break to around day 10 in the X-ray light curve. We show that for the physical parameters derived for GRB 070125, inverse Compton scattering effects are important throughout the afterglow evolution. While inverse Compton scattering does not affect radio and optical bands, it may be a promising candidate to delay the jet break in the X-ray band. Radio light curves show rapid flux variations, which are interpreted as due to interstellar scintillation, and are used to derive an upper limit of 2.4×10172.4 \times 10^{17} cm on the radius of the fireball in the lateral expansion phase of the jet. Radio light curves and spectra suggest a high synchrotron self absorption frequency indicative of the afterglow shock wave moving in a dense medium. Our broadband modeling favors a constant density profile for the circumburst medium over a wind-like profile (R2R^{-2}). However, keeping in mind the uncertainty of the parameters, it is difficult to unambiguously distinguish between the two density profiles.     Swab Materials and Bacillus anthracis Spore Recovery from Nonporous Surfaces

    Four swab materials were evaluated for their efficiency in recovery of Bacillus anthracis spores from steel coupons. Cotton, macrofoam, polyester, and rayon swabs were used to sample coupons inoculated with a spore suspension of known concentration. Three methods of processing for the removal of spores from the swabs (vortexing, sonication, or minimal agitation) and two swab preparations (premoistened and dry) were evaluated. Results indicated that premoistened swabs were more efficient at recovering spores than dry swabs (14.3% vs. 4.4%). Vortexing swabs for 2 min during processing resulted in superior extraction of spores when compared to sonicating them for 12 min or subjecting them to minimal agitation. Premoistened macrofoam and cotton swabs that were vortexed during processing recovered the greatest proportions of spores with a mean recovery of 43.6% (standard deviation [SD] 11.1%) and 41.7% (SD 14.6%), respectively. Premoistened and vortexed polyester and rayon swabs were less efficient, at 9.9% (SD 3.8%) and 11.5% (SD 7.9%), respectively

    An episomal DNA vector platform for the persistent genetic modification of pluripotent stem cells and their differentiated progeny

    The genetic modification of stem cells (SCs) is typically achieved using integrating vectors, whose potential integrative genotoxicity and propensity for epigenetic silencing during differentiation limit their application. The genetic modification of cells should provide sustainable levels of transgene expression, without compromising the viability of a cell or its progeny. We developed nonviral, nonintegrating, and autonomously replicating minimally sized DNA nanovectors to persistently genetically modify SCs and their differentiated progeny without causing any molecular or genetic damage. These DNA vectors are capable of efficiently modifying murine and human pluripotent SCs with minimal impact and without differentiation-mediated transgene silencing or vector loss. We demonstrate that these vectors remain episomal and provide robust and sustained transgene expression during self-renewal and targeted differentiation of SCs both in vitro and in vivo through embryogenesis and differentiation into adult tissues, without damaging their phenotypic characteristics

    Is There a Seamount Effect on Microbial Community Structure and Biomass? The Case Study of Seine and Sedlo Seamounts (Northeast Atlantic)

    Seamounts are considered to be “hotspots” of marine life but, their role in oceans primary productivity is still under discussion. We have studied the microbial community structure and biomass of the epipelagic zone (0–150 m) at two northeast Atlantic seamounts (Seine and Sedlo) and compared those with the surrounding ocean. Results from two cruises to Sedlo and three to Seine are presented. Main results show large temporal and spatial microbial community variability on both seamounts. Both Seine and Sedlo heterotrophic community (abundance and biomass) dominate during winter and summer months, representing 75% (Sedlo, July) to 86% (Seine, November) of the total plankton biomass. In Seine, during springtime the contribution to total plankton biomass is similar (47% autotrophic and 53% heterotrophic). Both seamounts present an autotrophic community structure dominated by small cells (nano and picophytoplankton). It is also during spring that a relatively important contribution (26%) of large cells to total autotrophic biomass is found. In some cases, a “seamount effect” is observed on Seine and Sedlo microbial community structure and biomass. In Seine this is only observed during spring through enhancement of large autotrophic cells at the summit and seamount stations. In Sedlo, and despite the observed low biomasses, some clear peaks of picoplankton at the summit or at stations within the seamount area are also observed during summer. Our results suggest that the dominance of heterotrophs is presumably related to the trapping effect of organic matter by seamounts. Nevertheless, the complex circulation around both seamounts with the presence of different sources of mesoscale variability (e.g. presence of meddies, intrusion of African upwelling water) may have contributed to the different patterns of distribution, abundances and also changes observed in the microbial community
